1. MacroGenics乳腺癌單抗新藥達(dá)到3期臨床終點(diǎn)
日前，專注于開發(fā)創(chuàng)新單克隆抗體癌癥療法的臨床階段生物醫(yī)藥公司MacroGenics宣布，其在研單抗藥物margetuximab，在治療轉(zhuǎn)移性人表皮生長(zhǎng)因子受體2陽性（HER2+）乳腺癌的關(guān)鍵3期試驗(yàn)SOPHIA中，取得了積極結(jié)果。MacroGenics計(jì)劃于2019年下半年為margetuximab提交生物制劑許可申請(qǐng)（BLA）。Margetuximab是MacroGenics公司研發(fā)的對(duì)抗體Fc端進(jìn)行優(yōu)化的HER2單克隆抗體。
2. 拜耳darolutamide組合療法3期結(jié)果公布，延長(zhǎng)前列腺癌無轉(zhuǎn)移生存期22個(gè)月
2月14日，治療非轉(zhuǎn)移性去勢(shì)抵抗性前列腺癌（nmCRPC）患者的關(guān)鍵性3期ARAMIS試驗(yàn)結(jié)果顯示，在研藥物darolutamide加雄激素剝奪療法（ADT），與安慰劑加ADT相比，統(tǒng)計(jì)顯著改善無轉(zhuǎn)移生存期（MFS），將腫瘤轉(zhuǎn)移或死亡風(fēng)險(xiǎn)降低59％。Darolutamide組的中位MFS為40.4個(gè)月，安慰劑組為18.4個(gè)月，中位MFS的總體差異為22個(gè)月。Darolutamide是一種在研非甾體雄激素受體拮抗劑，它能夠抑制雄激素受體功能和前列腺癌細(xì)胞的生長(zhǎng)。
3. 第一三共CSF1R抑制劑上市申請(qǐng)獲得FDA優(yōu)先審評(píng)
2月9日，第一三共（Daiichi Sankyo）公司宣布，該公司為CSF1R抑制劑pexidartinib遞交的新藥申請(qǐng)（NDA）已經(jīng)被美國(guó)FDA接受，同時(shí)FDA授予這一申請(qǐng)優(yōu)先審評(píng)資格。Pexidartinib用于治療腱鞘巨細(xì)胞瘤（TGCT），這是一種罕見的失能性腫瘤。如果獲得批準(zhǔn)，pexidartinib將成為治療TGCT的第一個(gè)也是唯一的獲批療法。
4. Trovagene公布o(jì)nvansertib與Zytiga聯(lián)合治療前列腺癌的2期研究進(jìn)展
Trovagene是一家臨床階段的腫瘤治療公司，采用精準(zhǔn)醫(yī)學(xué)方法開發(fā)針對(duì)細(xì)胞分裂（有絲分裂）的藥物，以治療白血病，淋巴瘤和實(shí)體腫瘤癌。2月14日，Trovagene公布了其正在進(jìn)行的2期研究進(jìn)展，該研究評(píng)估onvansertib聯(lián)合Zytiga（醋酸阿比特龍）/潑尼松治療轉(zhuǎn)移性去勢(shì)抵抗性前列腺癌（mCRPC）患者的療效和安全性。試驗(yàn)數(shù)據(jù)證明了onvansertib與Zytiga聯(lián)合治療的安全性和耐受性。Onvansertib是一種“first-in-class”的第三代口服和高選擇性絲氨酸/蘇氨酸polo樣激酶1（PLK 1）抑制劑，PLK酶在多種癌癥中過量表達(dá)，包括白血病，淋巴瘤和實(shí)體瘤。
